E6000 sticks to a lot of surfaces, but 'drying' depends on the surface chemistry. On uncoated cotton canvas, the adhesive can partially soak into the fabric, which helps the mechanical bond. On coated polyester or vinyl, the adhesive sits on top, so the bond is only as good as the surface preparation. Wipe it with isopropyl alcohol, and roughen it if possible.
I'm not 100% sure what your tote bag is made of, but check the label or the manufacturer's spec. If there's a waterproofing treatment, dry times can be longer. Don't assume cotton. Think about the edge of the tote (the arris) where fabric folds over the inner lining. That folded edge is often a different substrate than the main panel.
Step 2: Do the E6000 Arris test before you start
